There are all sorts of ways to calculate exactly what the DOF will be for any given shot at a given distance, focal length and aperture. But often you won't have the programs with you, unless you have it on a PDA like I do.

But you can use your camera's DOF preview (assuming it has one) and that will give you a pretty good guess. Also, just work with one of the calculators and play what if for a few moments and then in the future you'll have that information lodged in your mind for quick reference.
